• Opening Medley:
  • Million Dollar Quartet - Tony-winner Levi Kreis joined host Sean Hayes on the piano and performed "Blue Suede Shoes" with the company.
  • Memphis - Nominee Chad Kimball introduced the Opening Medley from his DJ booth.
  • Promises, Promises - Kristin Chenoweth sang "I Say a Little Prayer," accompanied by co-star and nominee Sean Hayes on the piano.
  • Come Fly Away - Nominee Karine Plantadit and the company danced to Frank Sinatra's "I'm Gonna Live Til I Die."
  • Everyday Rapture - Sherie Rene Scott and her back-up singers, the Mennonettes, went "Up the Ladder to the Roof."
  • Fela! -The company performed "Water No Get Enemy."
  • La Cage aux Folles - The Cagelles strutted to "I Am What I Am" as Sean Hayes accompanied them on piano.
  • American Idiot - The company sang "Boulevard of Broken Dreams," which was followed by the band Green Day playing "Holiday" and "Know Your Enemy."
• Million Dollar Quartet - The company sang a medley of "Down by the Riverside," "Let's Have a Party," and "Whole Lotta Shakin' Going On."
• La Cage aux Folles - Tony-winner Douglas Hodge, nominee Kelsey Grammer, and the cast enjoyed "The Best of Times."
• Memphis - Nominees Montego Glover and Chad Kimball led the cast in "Steal Your Rock 'n' Roll"
• A Little Night Music - Tony-winner Catherine Zeta-Jones sang "Send in the Clowns."
• Ragtime - Nominee Christiane Noll performed "Back to Before."
• Fela! - Nominees Sahr Ngaujah and Lillias White led the cast in a medley including "Everything Scatter," "Trouble Sleep," "Pipeline," I.T.T.," and "Yellow Fever."
• In a segment celebrating choreography, the cast of Come Fly Away danced to Sinatra's "That's Life"; and the Promises, Promises company featuring Sean Hayes performed the show's choreographed overture.
• Two stars of television's "Glee" paid tribute to their Broadway roots. Matthew Morrison sang "All I Need is the Girl" from Gypsy, and Lea Michele followed with "Don't Rain on My Parade" from Funny Girl.
• American Idiot - After being introduced by Green Day's Billie Joe Armstrong, the company performed the musical's title song.
• The Finale - The cast of the newly-designated Best Musical, Memphis, reprised "Steal Your Rock 'n' Roll."

Other Elements

Cast members from each of the four Best Play nominees presented brief synopses of the shows: Sarah Ruhl's In the Next Room or the vibrator play was introduced by Laura Benanti and Michael Cerveris; Geoffrey Nauffts's Next Fallby Patrick Breen and Patrick Heusinger; John Logan's Red by Alfred Molina and Eddie Redmayne; and Donald Margulies's Time Stands Still by Brian d'Arcy James and Laura Linney.

The productions nominated as Best Revival of a Play were also presented by their cast members: Fences - Viola Davis and Denzel Washington; Lend Me a Tenor - Justin Bartha, Anthony LaPaglia, and Tony Shalhoub; The Royal Family - Rosemary Harris and Jan Maxwell, A View from the Bridge - Scarlett Johansson and Liev Screiber.

2009 Tony-winners Gregory Jbara and Karen Olivo hosted the presentation of the InterContinental Hotels and Resorts Creative Arts Awards immediately before the telecast. This portion of the Tony Awards ceremony included 11 competitive awards, as well as the Regional Theatre Award, the Isabelle Stevenson Award, and Special Tony Awards for Lifetime Achievement in the Theatre.

The 2010 Tony Honors for Excellence in the Theatre were distributed at a special reception on the evening before the Tonys.

For the second time, fans in the New York area were able to watch the Tony Awards telecast right in the heart of Manhattan's Theatre District. Through a partnership between the Tonys, iXP Corporation, and the Times Square Alliance, the entire Tony Awards ceremony was shown live from Radio City Music Hall on the ClearChannel Spectacolor HD Screen in Times Square. The simulcast featured both the pre-telecast Creative Arts Awards and the Tony Awards broadcast, as well as NY1 News's coverage of the Audemars Piguet Tony Awards Red Carpet. Constantine Maroulis hosted the Times Square event, which included special appearances by Michelle Williams of Destiny's Child and Kerry Washington.
